Prednisone 20 mg, 500 Tablets Prednisone is used in the treatment of many disease processes including adrenal insufficiency, arthritis, lupus, allergies, asthma, dermatitis, blood and skin disorders. Prednisone 20 mg, 500 Tablets Prednisone is a synthetic corticosteroid used for many conditions. Its anti-inflammatory activity is approximately four times that of hydrocortisone. Corticosteroids are extremely effective anti-inflammatory drugs because they affect the inflammatory process at so many different levels. Prednisone is rapidly converted to prednisolone in the liver and in most instances, these drugs are considered to be roughly equivalent. Prednisone may be given by injection, orally or topically. Dogs and Cats: Prednisone is used for a wide variety of conditions in both dogs and cats. It may be used in emergency situations including, anaphylactic reactions, spinal chord trauma, and many forms of shock. It is used in the management and treatment of immune mediated disease such as immune mediated hemolytic anemia, or thombocytopenia: many CNS disorders: some neoplasia: dermatologic diseases: allergic reactions such as asthma, hives, and itching: inflammatory orthopedic diseases: endocrine disorders including Addison's: respiratory disease with an inflammatory component, inflammatory bowel diseases and many other conditions. Cats may require higher doses than dogs in order to achieve clinical response, but they are less likely to develop adverse side effects. Horses: Prednisone is given systemically to decrease inflammatory and immune responses. For years it was used orally to treat Chronic Obstructive Pulmonary Disease (COPD), and other allergic or immune-mediated disorders. Recent studies show that horses do not absorb oral prednisone, but they do absorb oral prednisolone. Other corticosteroids are preferred for intra-articular use. Side Effects: Systemic side effects to corticosteroids are generally dependent on dose and duration of treatment. Short-term use of prednisone is unlikely to cause adverse effects. Adverse effects are more common in animals on immunosuppressive doses. Side effects seen in dogs include polyuria, polydipsia, polyphagia, poor haircoat, GI disturbance, diarrhea, vomiting, weight gain, GI ulceration, pancreatitis, lipidemia, elevated liver enzymes, diabetes mellitus, muscle wasting, and possible behavioral changes. Polyuria, polydipsia, polyphagia may be seen in dogs even on short-term therapy. Although cats are less likely to develop side effects than dogs, occasionally polyuria, polydipsia, polyphagia, weight gain, GI disturbances and behavioral changes occur. Corticosteroids can cause or worsen gastric ulcers. Precautions: Chronic or inappropriate use of corticosteroids can cause life threatening hormonal and metabolic changes. Adverse effects due to corticosteroid treatment usually occur with long-term administration of the drug, especially when high doses are used. Alternate day therapy with short acting preparations is preferred. Animals who have received long-term therapy should be withdrawn slowly by tapering the dosage and prolonging the interval between doses. Corticosteroids suppress immune response. Animals receiving systemic corticosteroids may be more susceptible to bacterial or viral infections. Systemic corticosteroids can mask signs of infection, such as an elevated temperature. Systemic corticosteroids are contraindicated in patients with systemic fungal infections. (The treatment of Addison's disease may be considered an exception.) Animals in hepatic failure should receive prednisolone rather than prednisone. Corticosteroids should be avoided or used very carefully in young animals both because of immune suppression and the risk of GI ulcers. The structure is: Chemical Name: 4-Chloro-N-furfuryl-5-sulfamoylanthranillic acid. Furosemide is pharmaco-dynamically characterized by the following: 1) It is administered orally. The drug possesses diuretic activity either in the presence of acidosis or alkalosis. 1 2 4 5 6 INDICATIONS: DOGS - Furosemide Tablets are indicated for the treatment of edema (pulmonary congestion, ascites) associated with cardiac insufficiency and acute noninflammatory tissue edema. In cases of edema involving cardiac insufficiency, the continued use of heart stimulants such as digitalis or its glycosides is indicated. The rationale for efficacious use of diuretic therapy is determined by the clinical pathology producing the edema. CONTRAINDICATIONS: Animal reproductive studies have shown that furosemide may cause fetal abnormality and the drug is contraindicated in pregnant animals. Furosemide is contraindicated in anuria, furosemide hypersensitivity, hepatic coma, or during electrolytic imbalances. Monitor serum electrolytes, BUN and CO 2 frequently. Monitor serum potassium levels and watch for signs of hypocalcemia. Corticosteroids cause an additive potassium-depletion effect. PRECAUTIONS: Furosemide Tablets are a highly effective diuretic-saluretic which, if given in excessive amounts, may result in dehydration and electrolyte imbalance. Therefore, the dosage and schedule may have to be adjusted to the patient's need. The animal should be observed for early signs of electrolyte imbalance, and corrective measures administered. Early signs of electrolyte imbalance are increased thirst, lethargy, drowsiness or restlessness, fatigue, oliguria, gastro-intestinal disturbances and tachycardia. Special attention should be given to potassium levels. Furosemide Tablets may lower serum calcium levels and cause tetany in rare cases of animals having an existing hypocalcemic tendency. 7 8 9 10 11 Furosemide Tablets are contraindicated in anuria. Therapy should be discontinued in cases of progressive renal disease if increasing azotemia and oliguria occur during the treatment. Sudden alterations of fluid and electrolyte imbalance in an animal with cirrhosis may precipitate hepatic coma, therefore, observation during period of therapy is necessary. DESCRIPTION: Prednisolone, like methylprednisolone, is a potent anti-inflammatory steroid. Prednisolone, 11,17,21-trihydroxypregna-1,4-diene-3,20-dione, is a synthetic dehydrogenated analogue of cortisone. Prednisolone and methylprednisolone have a greater anti-inflammatory potency and less tendency to induce sodium and water retention than the older corticoids, cortisone and hydrocortisone. The relative anti-inflammatory potency for hydrocortisone is 1.0; cortisone is 0.8; prednisolone is 4 and methylprednisolone is 5. The relative sodium retaining potency for hydrocortisone is 4; prednisolone is 3 and methylprednisolone is 2. 1,2 INDICATIONS: PrednisTab is intended for oral use in dogs. The indications for PrednisTab are the same as those for other anti-inflammatory steroids and comprise the various collagen, dermal, allergic, ocular, otic, and musculoskeletal conditions known to be responsive to the anti-inflammatory corticosteroids. Representative of the conditions in which the use of steroid therapy and the benefits to be derived therefrom have had repeated confirmation in the veterinary literature are: (1) dermal conditions, such as nonspecific eczema, summer dermatitis, and burns; (2 allergic manifestations, such as acute urticaria, allergic dermatitis, drug and serum reactions, bronchial asthma, and pollen sensitivities; (3) ocular conditions, such as iritis, iridocyclitis, secondary glaucoma, uveitis, and chorioretinitis; (4) otic conditions, such as otitis externa; (5) musculoskeletal conditions, such as myositis, rheumatoid arthritis, osteoarthritis, and bursitis; (6) various chronic or recurrent diseases of unknown etiology such as ulcerative colitis and nephrosis. In acute adrenal insufficiency, prednisolone may be effective because of its ability to correct the defect in carbohydrate metabolism and relieve the impaired diuretic response to water, characteristic of primary or secondary adrenal insufficiency. However, because this agent lacks significant mineralocorticoid activity, hydrocortisone sodium succinate, hydrocortisone, or cortisone should be used when salt retention is indicated. CONTRAINDICATIONS: Do not use in viral infections. Prednisolone, like methylprednisolone, is contraindicated in animals with peptic ulcer, corneal ulcer, and Cushingoid syndrome. The presence of diabetes, osteoporosis, predisposition to thrombophlebitis, hypertension, congestive heart failure, renal insufficiency, and active tuberculosis necessitates carefully controlled use. Some of the above conditions occur only rarely in dogs, but should be kept in mind. CAUTION: Because of its inhibitory effect on fibroplasia, prednisolone may mask the signs of infection and enhance dissemination of the infecting organism. Hence, all animal patients receiving prednisolone should be watched for any evidence of intercurrent infection. Should infection occur, it must be brought under control by use of appropriate antibacterial measures, or administration of prednisolone should be discontinued. Warning: Not for human use. Clinical and experimental data have demonstrated that corticosteroids administered orally or by injection to animals may induce the first stage of parturition if used during the last trimester of pregnancy and may precipitate premature parturition followed by dystocia, fetal death, retained placenta, and metritis. Denosyl is available in three strengths: as an enteric-coated 90 mg, 225 mg, and 425 mg tablet of active S-Adenosylmethionine. Because S-Adenosylmethionine is extremely sensitive to moisture, tablets are individually sealed in specialized moisture-resistant blister packs. Background S-Adenosylmethionine is an endogenous molecule synthesized by cells throughout the body and is formed from the amino acid methionine and ATP. It is an essential part of three major biochemical pathways: transmethylation, transsulfuration and aminopropylation. As part of these pathways, S-Adenosylmethionine is essential to all cells and is particularly important in hepatocytes because of their central role in metabolism. A deficiency of S-Adenosylmethionine, therefore, may initiate or contribute to abnormalities of cellular structure and function in the liver as well as many other body tissues. Conversely, exogenous administration of S-Adenosylmethionine has been shown to result in improvements in hepatocellular function in both in vivo and in vitro studies, without cytotoxicity or significant side effects. 1-9 Precursors of S-Adenosylmethionine do not have similar effects. Administration of methionine to animals with decreased liver function may not increase hepatic S-Adenosylmethionine levels and may be toxic. 6 The best way to increase S-Adenosylmethionine levels in the body is by direct supplementation with S-Adenosylmethionine. Purpose Denosyl has been shown to increase hepatic glutathione levels in cats and dogs. 1,7 Glutathione is a potent antioxidant that protects hepatic cells from toxins and death. A study found that low liver glutathione concentrations are common in dogs and cats with decreased hepatobiliary function. 10 Denosyl is recommended to improve hepatic glutathione levels in patients to help maintain and protect liver function. Denosyl may also be used in other areas of tissue oxidant injury and RBC fragility caused by certain toxins or drugs which are related to reduced glutathione concentrations. 6 Denosyl, however, goes beyond increasing glutathione levels and has been shown to protect liver cells from cell death 2,8 and may be useful in cell regeneration. 6 A study has also shown that Denosyl may improve bile flow in cats. 5 Pharmacokinetics In a pharmacokinetic study with fasted dogs given Denosyl, peak plasma levels were recorded within 4 hours of administration. 1 A pharmacokinetic study in fasted cats showed peak plasma levels at 2-4 hours post Denosyl administration. 7,11 Safety Denosyl demonstrates an exceptionally wide margin of safety. Oral acute toxicity studies in rats indicated an LD50 greater than 4,640 mg/kg. 6 Clinically healthy dogs administered 20 mg/kg/day of Denosyl for 6 weeks and clinically healthy cats administered Denosyl at 2 times the recommended daily amount for 113 days remained healthy with no adverse effects from administration of Denosyl. 1,7 Administration as a Nutritional Supplement Because the tablet must be kept intact to prevent destruction by stomach acid, administration range is based on the nearest whole-tablet. The chart below is provided as a guide for administration. Daily administration may also be calculated based on 20 mg/kg of body weight and rounded to the closest tablet size or combination of sizes. For optimal absorption, tablets should be given on an empty stomach, at least one hour before feeding, as the presence of food decreases the absorption of S-
